ADAM18

ADAM18: A Critical Protein in Immune Function and Disease

Description:

ADAM18, also known as disintegrin and metallopeptidase domain-18, is a transmembrane protein that plays a crucial role in cell-cell interactions and proteolytic processing. It is expressed on the surface of various immune cells, including macrophages, dendritic cells, and neutrophils.

ADAM18 consists of an extracellular domain with a disintegrin-like domain, a metalloprotease domain, and a cysteine-rich domain. The disintegrin-like domain facilitates cell adhesion, while the metalloprotease domain cleaves and activates a wide range of substrates.

Associated Diseases:

Dysregulation of ADAM18 has been implicated in the pathogenesis of several diseases, including:

  • Cardiovascular disease: ADAM18 promotes the shedding of pro-atherogenic factors, contributing to plaque formation and thrombosis.
  • Osteoarthritis: ADAM18 is involved in the degradation of cartilage matrix components, leading to cartilage destruction and joint pain.
  • Cancer: ADAM18 facilitates tumor cell migration, invasion, and metastasis.

Did you Know ?

  • ADAM18 deficiency in mice results in severe immune dysfunction, highlighting its essential role in immune responses.
