A disintegrin and metalloproteinase with thrombospondin motifs 4 (ADAMTS4), also known as aggrecanase-1, is a crucial enzyme involved in various physiological and pathological processes. It belongs to the ADAMTS family, a group of zinc-dependent metalloproteases that play significant roles in extracellular matrix (ECM) remodeling. ADAMTS4 specifically cleaves aggrecan, a major proteoglycan component of cartilage, and is essential for cartilage homeostasis and development.

Associated Diseases

Dysregulation of ADAMTS4 activity has been implicated in several human diseases, including:

  • Osteoarthritis (OA): In OA, increased ADAMTS4 activity leads to the degradation of aggrecan, disrupting cartilage structure and function. This contributes to the progressive loss of cartilage and joint pain associated with OA.

  • Rheumatoid arthritis (RA): In RA, ADAMTS4 is involved in the breakdown of synovial tissue, a membrane that lines the joints. Excessive ADAMTS4 activity exacerbates cartilage damage and promotes joint inflammation.

  • Cancer: ADAMTS4 has been found to play a role in tumor progression and metastasis. It can cleave ECM components, facilitating tumor cell invasion and spread.
