Description

The ALPK1 (alpha kinase 1) is a protein-coding gene located on chromosome 4.

Alpha-protein kinase 1 is an enzyme encoded by the ALPK1 gene in humans. Unlike most eukaryotic kinases, alpha kinases, such as ALPK1, recognize phosphorylation sites in peptides with an alpha-helical conformation.

ALPK1 is a serine/threonine kinase that plays a critical role in innate immunity by detecting bacterial pathogen-associated molecular pattern metabolites (PAMPs) and initiating an immune response. It specifically recognizes and binds ADP-D-glycero-beta-D-manno-heptose (ADP-Heptose), a PAMP found in Gram-negative and some Gram-positive bacteria. Binding of ADP-Heptose stimulates ALPK1's kinase activity, leading to phosphorylation and activation of TIFA, which triggers pro-inflammatory NF-kappa-B signaling. Additionally, ALPK1 may be involved in monosodium urate monohydrate (MSU)-induced inflammation by phosphorylating unconventional myosin MYO9A. ALPK1 may also play a role in apical protein transport by phosphorylating unconventional myosin MYO1A and may contribute to ciliogenesis.
