The ARID4B (AT-rich interaction domain 4B) is a protein-coding gene located on chromosome 1.

ARID4B (AT-rich interactive domain-containing protein 4B) is a protein encoded by the ARID4B gene in humans. It shares sequence similarity with retinoblastoma-binding protein-1 and is a subunit of the histone deacetylase-dependent SIN3A transcriptional corepressor complex. This complex plays a role in various cellular processes including proliferation, differentiation, apoptosis, oncogenesis, and cell fate determination. ARID4B has been identified as a molecular marker associated with a range of human malignancies. Several different isoforms of ARID4B exist due to alternative splicing.

ARID4B acts as a transcriptional repressor, potentially contributing to the assembly and enzymatic activity of the Sin3A corepressor complex, or mediating interactions between this complex and other regulatory complexes. It plays a role in epigenetic modifications at the PWS/AS imprinting center, potentially functioning in a complex with RB1 and ARID4A. In spermatogenesis, it collaborates with ARID4A to act as a transcriptional coactivator for the androgen receptor (AR), enhancing the expression of genes vital for sperm maturation. It regulates the expression of CLDN3, a tight junction protein in the testis, crucial for maintaining the blood-testis barrier. ARID4B is involved in myeloid homeostasis, regulating histone methylation in bone marrow cells and influencing the expression of genes involved in hematopoiesis. It might function as a leukemia suppressor. It is part of a Sin3A corepressor complex, alongside SIN3A, SAP130, SUDS3/SAP45, SAP180, HDAC1, and HDAC2. It interacts with ARID4A and AR.
