Description

The CLNK (cytokine dependent hematopoietic cell linker) is a protein-coding gene located on chromosome 4.

CLNK is a crucial adapter protein that plays a vital role in regulating immune receptor signaling. It influences B-cell receptor signaling through PLC-gamma and FC-epsilon R1-mediated mast cell degranulation. In collaboration with FGR, CLNK acts as a negative regulator of natural killer cell-activating receptors and inhibits interferon-gamma production. Additionally, CLNK positively regulates both T-cell receptor and natural killer T cell receptor signaling in CD4-positive NKT cells. Alongside MAP4K1, it enhances CD3-triggered activation of T-cells and subsequent IL2 production. CLNK may also participate in tumor necrosis factor induced cell death by promoting reactive oxidative species generation and MLKL oligomerization, leading to necrosis. It plays a role in the phosphorylation of LAT and may be involved in high-affinity immunoglobulin epsilon receptor signaling in mast cells.
