HMCES acts as a sensor of abasic sites (missing bases) in single-stranded DNA (ssDNA), a crucial role in maintaining genomic integrity. It promotes error-free repair by forming a covalent cross-link with the abasic site, preventing error-prone repair mechanisms. This cross-link can be reversed or degraded, allowing for accurate repair. HMCES also plays a protective role in B-cell development, limiting deletions during somatic hypermutation of immunoglobulin genes and contributing to class switch recombination.
