IL-24 is a multifunctional cytokine primarily produced by T-cells. It plays a regulatory role in immune responses, tissue homeostasis, host defense, and oncogenesis. IL-24 exhibits antiviral properties and triggers type I interferon responses during influenza infections. It signals through two receptor complexes: IL20RA/IL20RB or IL20RB/IL22RA1, activating the JAK1-STAT3 and MAPK pathways, and promoting the secretion of pro-inflammatory mediators such as IL8 and MMP1. IL-24 also maintains endoplasmic reticulum homeostasis by restricting eIF2alpha-CHOP pathway-mediated stress signals. Additionally, it acts as a quality control mechanism for the ubiquitin proteasome system, alerting the cell to proteasome dysfunction through the activation of PKR/EIF2AK2.
