Leucine-rich repeat containing 18 (lrrc18) is a gene that encodes a protein essential for maintaining cellular function and structure. It plays a crucial role in various cellular processes, including neurite growth, axonal transport, autophagy, and vesicular trafficking.

The lrrc18 protein is composed of several key domains, including:

  • Leucine-rich repeat (LRR) domain: Involved in protein-protein interactions and potentially mediates the interaction with membrane receptors.
  • GTPase domain: Regulates GTP hydrolysis, which is critical for energy metabolism and signaling pathways.
  • WD40 domain: Facilitates protein-protein interactions and may be involved in the formation of protein complexes.

Associated Diseases

lrrc18 mutations and dysregulation have been linked to several neurodegenerative diseases, including:

  • Parkinson's disease (PD): Mutations in lrrc18 are the most common genetic cause of familial PD. These mutations lead to abnormal accumulation of the lrrc18 protein, resulting in neuronal dysfunction and neurodegeneration.
  • Amyotrophic lateral sclerosis (ALS): lrrc18 mutations have also been identified in patients with ALS. These mutations affect the GTPase domain and impair lrrc18's function in axonal transport.
  • Charcot-Marie-Tooth disease type 2 (CMT2): Mutations in lrrc18 have been linked to CMT2, a peripheral neuropathy characterized by progressive weakness and muscle atrophy.
  • Mitochondrial disorders: lrrc18 has been implicated in mitochondrial function, and mutations in lrrc18 have been associated with mitochondrial defects and oxidative stress.
