Description

The NEIL2 (nei like DNA glycosylase 2) is a protein-coding gene located on chromosome 8.

Endonuclease VIII-like 2 is an enzyme encoded by the NEIL2 gene in humans. It belongs to a group of DNA glycosylases related to the bacterial Fpg/Nei family.

NEIL2 participates in the base excision repair (BER) pathway, which removes damaged or modified bases from DNA. It exhibits DNA glycosylase activity against 5-hydroxyuracil and other oxidized cytosine derivatives, particularly in mismatched double-stranded DNA regions (DNA bubbles). However, its activity towards thymine glycol, 2-hydroxyadenine, hypoxanthine, and 8-oxoguanine is limited or absent. NEIL2 also possesses AP (apurinic/apyrimidinic) lyase activity, enabling it to introduce nicks in the DNA strand. It cleaves the DNA backbone through beta-delta elimination, generating a single-strand break at the site of the removed base with both 3'- and 5'-phosphates.
