The PARD6B (par-6 family cell polarity regulator beta) is a protein-coding gene located on chromosome 20.

PARD6B, or Partitioning defective 6 homolog beta, is a protein encoded by the PARD6B gene in humans. It belongs to the PAR6 family and contains a PSD95/Discs-large/ZO1 (PDZ) domain, an OPR domain, and a semi-Cdc42/Rac interactive binding (CRIB) domain. This cytoplasmic protein is involved in asymmetrical cell division and cell polarization processes, working as part of a multi-protein complex.

PARD6B, also known as PAR-6 beta, is an adapter protein crucial for asymmetrical cell division and cell polarization. It plays a role in forming epithelial tight junctions, likely by preventing PARD3 from interacting with F11R/JAM1, which inhibits tight junction assembly. The PARD6-PARD3 complex connects GTP-bound Rho small GTPases to atypical protein kinase C proteins.
