Description

The PCID2 (PCI domain containing 2) is a protein-coding gene located on chromosome 13.

PCID2 is a protein encoded by the PCID2 gene in humans. It is a component of the TREX-2 complex, which regulates mRNA export from the nucleus. PCID2 regulates the expression of MAD2L1, a cell division checkpoint protein, and interacts with and stabilizes BRCA2 protein. Alternative splicing produces multiple transcript variants.

PCID2 plays a crucial role in B-cell survival by regulating the expression of the cell cycle checkpoint protein MAD2L1 during B cell differentiation. As part of the TREX-2 complex, it facilitates the export of mRNAs from the nucleus to the cytoplasm. PCID2 also binds and stabilizes the BRCA2 protein, contributing to the control of R-loop-associated DNA damage and transcription-associated genomic instability. Furthermore, PCID2 interacts with ZNHIT1, a component of the SRCAP chromatin remodeling complex, inhibiting its activity. This prevents the deposition of histone variant H2AZ1/H2A.Z at the nucleosomes of key lymphoid fate regulator genes, suppressing their expression and restricting lymphoid lineage commitment.
