Description

The UBA2 (ubiquitin like modifier activating enzyme 2) is a protein-coding gene located on chromosome 19.

UBA2, also known as SUMO-activating enzyme subunit 2 (SAE2), is a human gene that encodes an enzyme involved in protein sumoylation, a process that regulates protein structure and localization. UBA2 forms a heterodimer with SAE1, acting as a SUMO-activating enzyme. The UBA2 cDNA spans 2683 bp, encoding a 640-amino acid peptide with a molecular weight of 72 kDa. It is comprised of three domains: an adenylation domain, a catalytic Cys domain, and a ubiquitin-like domain. SUMO-1 binds to UBA2 between the catalytic Cys domain and the UbL domain. UBA2, along with SAE1, catalyzes the activation of SUMO-1 and transfers it to Ubc9, the only known E2 enzyme in sumoylation.

UBA2, also known as SAE2, forms a heterodimer with SAE1 to act as an E1 ligase for SUMO1, SUMO2, SUMO3, and likely SUMO4. This heterodimer utilizes ATP to activate SUMO proteins, subsequently forming a thioester bond between a SUMO protein and a conserved cysteine residue within UBA2/SAE2.
