Description

The FBXO45 (F-box protein 45) is a protein-coding gene located on chromosome 3.

FBXO45 is a human gene that encodes a protein with an F-box domain and a SPRY domain. The protein is involved in the ubiquitin-proteasome system, which is responsible for the degradation of proteins in cells.

FBXO45 is a component of the E3 ubiquitin ligase complex, which also includes MYCBP2 and SKP1. It plays a role in both substrate recognition and complex assembly. FBXO45 is involved in various cellular processes, including:

  • Neuromuscular synaptogenesis: essential for normal development of connections between neurons and muscles.
  • Axon pathfinding: guides the growth of axons, which are the long projections of neurons that transmit signals.
  • Neuronal migration: regulates the movement of neurons during brain development through interaction with N-cadherin/CDH2.
  • Neurotransmission: contributes to the regulation of communication between neurons at mature synapses.
  • Control of synaptic activity: may regulate synaptic activity by controlling UNC13A through a ubiquitin-dependent pathway.
  • Ubiquitination and degradation of specific proteins:
    • Promotes the degradation of TP73, a transcription factor.
    • Polyubiquitinates NMNAT2, an axon maintenance factor, and regulates its stability.
    • Ubiquitinates FBXW7 during prolonged mitotic arrest, leading to its degradation and preventing mitotic cell death.
  • Response to influenza infection: mediates the degradation of interferon-lambda receptor IFNLR1 by the ubiquitin-proteasome system.
