Description

The TBC1D7 (TBC1 domain family member 7) is a protein-coding gene located on chromosome 6.

TBC1D7, also known as Cell migration-inducing protein 23, is a non-catalytic component of the TSC-TBC complex. This complex acts as a negative regulator of the mTORC1 complex, a central nutrient sensor that promotes growth by stimulating anabolic reactions and macromolecule biosynthesis. The TSC-TBC complex functions as a GTPase-activating protein (GAP) for the small GTPase RHEB, a direct activator of mTORC1's protein kinase activity. When nutrients are scarce, the TSC-TBC complex inhibits mTORC1, preventing the phosphorylation of ribosomal protein S6 kinase (RPS6KB1 and RPS6KB2) and EIF4EBP1 (4E-BP1) by mTORC1 signaling. Upon nutrient availability, the TSC-TBC complex is inactivated, relieving the inhibition of mTORC1.
