Description

The GIGYF2 (GRB10 interacting GYF protein 2) is a protein-coding gene located on chromosome 2.

GIGYF2 is a key component of the 4EHP-GYF2 complex, a multiprotein complex that acts as a repressor of translation initiation. GIGYF2 functions as a bridge between EIF4E2 and ZFP36/TTP within this complex, linking translation repression with mRNA decay. It also recruits and facilitates the association of the 4EHP complex with DDX6, a decapping effector protein essential for ZFP36/TTP-mediated downregulation of AU-rich mRNA. GIGYF2 may cooperate with GRB10 to regulate tyrosine kinase receptor signaling, particularly for IGF1 and insulin receptors. In collaboration with EIF4E2, GIGYF2 participates in ribosome-associated quality control (RQC) by sequestering the mRNA cap, preventing ribosome initiation, and reducing the translational burden on problematic messages. This process operates in parallel to RQC-mediated degradation of stalled nascent polypeptides. GIGYF2 and EIF4E2 function downstream and independently of ZNF598, which acts as a scaffold for their recruitment to faulty mRNA, although alternative mechanisms may exist.
