Description

The RWDD3 (RWD domain containing 3) is a protein-coding gene located on chromosome 1.

RWD domain-containing protein 3 is a protein that in humans is encoded by the RWDD3 gene.

RWDD3 is an enhancer of SUMO conjugation. It interacts with UBE2I/UBC9 to promote SUMO conjugation to proteins. This process involves promoting the binding of E1 and E2 enzymes, facilitating thioester linkage between SUMO and UBE2I/UBC9, and transferring SUMO to target proteins such as HIF1A, PIAS, NFKBIA, NR3C1, and TOP1. RWDD3's isoforms 1 and 2 positively regulate the NF-kappa-B signaling pathway by enhancing the sumoylation of NFKBIA, which stabilizes NFKBIA and increases inhibition of NF-kappa-B transcriptional activity. These isoforms also negatively regulate the hypoxia-inducible factor-1 alpha (HIF1A) signaling pathway by promoting HIF1A sumoylation, leading to HIF1A stabilization, transcriptional activity, and increased expression of its target gene VEGFA during hypoxia. Isoform 2 further promotes sumoylation and transcriptional activity of the glucocorticoid receptor NR3C1 and strengthens the interaction between SUMO1 and NR3C1 with UBE2I/UBC9. Notably, RWDD3 does not affect ubiquitination.
