Ubiquitin specific peptidase 22 is a protein that in humans is encoded by the USP22 gene on chromosome 17.

USP22 is a histone deubiquitinating enzyme that is part of the SAGA complex, a transcription regulatory histone acetylation complex. It catalyzes the deubiquitination of histones H2A and H2B, acting as a coactivator. USP22 is recruited to specific gene promoters by activators like MYC, where it is essential for transcription. It is also crucial for nuclear receptor-mediated transactivation and cell cycle progression.

USP22 is also known as USP3L.
