ZNRF3
Description
The ZNRF3 (zinc and ring finger 3) is a protein-coding gene located on chromosome 22.
ZNRF3 is an E3 ubiquitin-protein ligase that negatively regulates the Wnt signaling pathway by ubiquitinating and degrading Frizzled and LRP6, components of the Wnt receptor complex. It acts on both canonical and non-canonical Wnt signaling pathways. ZNRF3 acts as a tumor suppressor in the intestinal stem cell zone by inhibiting Wnt signaling, thereby limiting the size of this zone. Along with RSPO2 and RNF43, it forms a master switch that governs limb specification.
